Activating reagent for carboxylic acids

N-hydroxysuccinimide (HOSu) finds its most significant application in the formation of isolable activated derivatives of Nα-protected α-amino acids. These derivatives then readily participate in smooth coupling reactions with amino esters. The typical method involves using 1,3-Dicyclohexylcarbodiimide (DCC) as the initial coupling agent (the HOSu-DCC method). Alternatively, N-hydroxyphthalimide (NHPI) can be used similarly. However, HOSu holds a distinct advantage due to its high water solubility, which greatly facilitates the purification of the reaction products. This water solubility allows for easy separation and purification of the activated derivatives, making HOSu an advantageous choice for such coupling reactions. Of course, the HOSu–DCC method is not limited to peptide synthesis and is well suited to the preparation of amides in general, both from ammonia and primary amines.
